To employ this technique as routine quality control test in vaccine industry, validation of this method according to regulatory guidelines was necessary. In present study to validate this method and to generate reliable data, various experiments were designed for execution of different validation parameters.In the present work, the polysaccharide estimation using anthrone reagent has been found quite easier and reproducible. Range of linearity was observed to be 5-125 µg with correlation coefficient 0.999. Anthrone assay for determination of polysaccharide was found to be specific, precise, accurate, robust and stable. First, a screening assay was developed to evaluate virus inactivation and preservation of the virion structure. Second, possible vaccine virus adaptation problems to a cell line are solved. This research is based on porcine reproductive and respiratory syndrome virus (PRRSV), but can be valuable for development of other vaccines as well.PRRSV leads to reproduction failure in sows and is associated with the respiratory disease complex in pigs. Although commercial vaccines exist, PRRSV is still difficult to control in the field. This can be due to modification of epitopes during the inactivation procedure or virus adaptation to a cell line.We developed a new inactivated PRRSV vaccine with virus grown on a PRRSV sensitive pig cell line to minimize mutations in viral structural proteins. Furthermore, we screened different inactivation methods for their ability to preserve viral entry-associated domains. Neuware -Vaccine development against blood-stage antigens is aimed at inducing inhibitory antibodies towards the circulating merozoites, thus preventing them from infecting red blood cells. One such blood stage candidate is the 19 kDa C-terminus of merozoite surface protein-1 (MSP-1C). MSP-1C has been shown to induce both inhibitory as well as blocking antibodies, the latter blocking the protective effects of the former. Inhibitory antibodies will bind to MSP-1C and inhibit merozoite invasion of red blood cell but the binding of blocking antibodies can prevent binding of inhibitory antibodies thereby allowing the parasite to invade an erythrocyte. This book provides some information regarding the development of blood-stage malaria vaccine based on mutated version of MSP-1C which induces inhibitory antibodies against malarial parasite. The use of live recombinant Mycobacterium bovis bacille Calmette Guerin as a vaccine vehicle was also described in this book. Neuware -MIS416 is a novel vaccine adjuvant generated from propionibacterium acnes cell wall skeletons comprising naturally occurring ligands with the ability to induce effective adaptive immunity. In this research, I aimed to enhance the therapeutic potential of MIS416-adjuvanted vaccines by directly conjugating specific peptides derived from Tumors Associated Antigens (TAA) and siRNAs, achieving co-delivery of antigen and adjuvant. Using the high-affintiy streptavidin-biotin complex as a conjugation strategy for peptide attachment, we were able to irreversibly couple different fluorescent molecules to the surface of MIS416 as proof of principle. The OVA-derived peptide SIINFEKL, was then coupled to MIS416 and this MIS416-peptide conjugate was pulsed onto Dendritic cells (DC) showing that DC were activated by the conjugate. Flow cytometry analysis showed that levels of MHCII, CD40, CD80 and CD86 were upregulated by our novel vaccine conjugate.
