Resulting in initiation and propagation reactive oxygen species which attacking macromolecules of cells such as proteins, lipids, RNA and DNA and ultimately cells death. Leading to a state of oxidative stress associated with many of health problems such as heart failure, liver cirrhosis, fibrosis, gallbladder disorders, diabetes, arthritis, infertility, and cancer. The available chelators drugs can forming soluble, stable complexes with excessive iron andexcreted it in the feces and/or urine. But due to, their high cost, severe side effects, poor oralbioavailability or short plasma half-life make them suboptimal.Sixteen plant leaves were collected, authenticated and subjected to exhausted extraction by 70% methanol. The obtained leaves extracts were screened for their iron chelation activity using 2,2`- bipyridyl assay. Among the screened extracts, the Mangiferaindica leaves showed the highest iron chelation activity(69.71 ± 0.27%)comparable to positive control, EDTA(70.30 ± 0.08%).The total extract of theM. indica leaves was partitioned with different solvents (petroleum ether, methylene chloride, ethyl acetate and n-butanol). The resultingfractions were screened for their iron chelation activity using 2, 2`- bipyridyl assay.EtOAcfractionhad the significant chelating activity (123 ± 2.75%),more potent than EDTA(64.12 ± 1.48%) and total extract of M. indica leaves (69.71 ± 0.27%).The mangiferin compound was subjected to comparison with desferal® to assurance the quality of its chelating activity throughdetermine the active sites responsible of its chelating property and evaluation the effect of its iron-stable complex on its antioxidant activity.According to introduced results, through different biological analysis. We found that, the iron-overloaded rats treated with ethyl acetate fraction had significant decreases in iron accumulation within liver and spleen (the major organs affected by iron overload). We assume that, the mangiferin may protect from reactive oxygenspecies-stimulatedseveral of disordersby its iron chelatingactivity which promotes its antioxidant property.

These plant were extracted and screened for their activity towards green synthesis of AgNPs. A new modified method was used in order to screen large number of extracts in short time (high throughput screening). In this method, the synthesized nanoparticles were evaluated in using 96-well plate which is transparent to facilitate the color monitoring and easy to be read spectrophotometrically using microplate reader. The green synthesis activity by the different plant extracts was monitored and evaluated by color change and UV-Vis absorption at time intervals.Out of these 42 plants, only 3 plants viz.,Pyllanthusemblicafruits, Psidium guajava leaves and Lawsoniainermisleaves, showed high AgNPs synthetic activity. The most active plant extracts were further partitioned with ethyl acetate. The results showed that the EtOAc fraction of the three active plants showed superior AgNPs green synthesis over their remaining aqueous. Thus, chromatographic columns were used to isolate the active compounds from the EtOAc fraction of each plant.Metal nanoparticles synthesis is a leading topic of research in modern material science owing to their distinctive potential applications in the field of electronic, optoelectronic, information storage and Health care. Druck auf Anfrage Neuware - Printed after ordering - Akademische Arbeit aus dem Jahr 2013 im Fachbereich Chemie - Biochemie, , Sprache: Deutsch, Abstract: In this article we report the isolation, characterization and evaluation of in vitro anti-colon cancer activity of a new gingerol derivative, namely (S,E)-5-hydroxy-1-(4-hydroxy-3- methoxyphenyl)tetradec-8-en-3-one, (N6), and a known sesquiterpene (1,2- Dihydroxybisabola-3,10-diene, N7), isolated from Zingiber officinale rhizomes for the first time, together with other five known compounds. 6-gingerol (N1), 8-gingerol (N2), 10- gingerol (N3), 4-gingerol (N4) and 4`-O-methyl-6-gingerol (N5).The isolated compounds were identified using different spectroscopic techniques (1D and 2D-NMR, MS and IR). A modified chromatographic method was established and optimized to isolate the major gingerols (N1, N2, and N3) in grams. Thereafter the cytotoxic activity of the isolated compounds was evaluated against colorectal carcinoma cell line, Caco-2. It was observed that anti-colon cancer activity of N2 was superior to that of 5-FU, the classic reference cytotoxic agent. Additionally, the length of the aliphatic chain in 8-gingerol (N2) is optimum for the anticancer activity and any decrease, as in N1 and N4, or increase, as in N3, in the side chain length leads to gradient decrease in the cytotoxic activity.Following this further, methylation of phenolic OH group (N5), leads to a dramatic decrease in the anticancer activity. Furthermore, loss of aromaticity (N7), results in complete loss of the cytotoxic activity. On the other hand, introducing a pi-bond in the aliphatic side chain enhances the anti-cancer activity as depicted in N6. Taken together, the aromaticity, the side chain length as well as the presence of free phenolic group contribute significantly to the anti-colon cancer activity of gingerol and its derivatives. These results open up a new window in the rational design of gingerol-based semisynthetic drugs with improved anticolon cancer activity.
