Highly critical for the proper function of lysosomes, endosomes, and lysosome-related organelles is the tight regulation of various fusion and fission processes and the regulation of proton and other ionic concentrations within the endolysosomal system. Calcium permeable, non-selective cation channels of the TRP (transient receptor potential) superfamily, namely TRPML channels (mucolipins) and TPCs (two-pore channels) have been found to play important roles in these processes. This thesis provides insights into the function and physiology of TRPML and TPC channels, explains the molecular basis of pathologies associated with diseases caused by loss or mutation of these channels, and discusses potential therapeutic approaches.…

This seminal discovery has opened up a 'hot' new field of basic research and launched drug discovery efforts into the large family (by the latest count, 28 mammalian members and 27 in humans) of TRP ion channels. Indeed, it took less than a decade for the first potent, small molecule TRPV1 antagonists to enter phase 1 clinical trials, closely followed by TRPA1 and TRPM8 antagonists. The literature on TRP channels is immense. TRPV1 alone is a keyword in over 5000 publications searchable in PubMed. Clearly, it is not possible to capture the entire literature in a single thematic issue. Consequently, the selection of articles presented in this book represents a sampling of the literature, and is admittedly subjective. We tried to survey the wide range of human diseases in which TRP channels have been implicated, ranging from chronic pain through asthma and diabetes to cancer, and highlight the channels that appear to hold the greatest promise for therapeutic targeting. TRPC proteins belong to the TRP channel superfamily, which has around 30 members. Just like TRP channels, TRPCs are non-selectively permeable to cations, with a selectivity of calcium over sodium that varies among different members. The TRPC subfamily consists of six members, grouped on the basis of the similarities in genesequence and proteinstructure in mammalians. What sets TRPCs apart from other subfamilies in TRPs is that their activation, stimulated by a membrane receptor-phospholipid C (PLC) cascade, contributes to the slow and sustained elevation of intracellular free calcium. Calcium ions, one of the most important types of second messengers, mediate a variety of physiological functions in the brain, including progenitor cell proliferation, dendritic formation, synaptic transmission and neuronal survival.
