We plan to continue the tradition of providing a forum for exten sive, critical reviews of individual virus groups, as exemplified by the present volume. But th…e pace of discovery is accelerating so rapidly that we feel the need to offer an additional format: volumes that contain collections of shorter, topical reviews on a group of related subjects. Such collections might cut across con ventional boundaries between virus groups, dealing, as an example, with a partic ular aspect of virus-cell interaction. Admittedly, this new format stretches the term 'monograph' beyond the accepted definition, but we believe that we should pay that price to maintain the usefulness of the series as a medium of scientific communication. Whenever possible, we will enlist the aid of deputy editors to bring such col lections to fruition. As in the past, the editors and the publisher will welcome suggestions for topics and contributions.

The first chapter, by Drs. Condit and Niles, discusses poxvirus genetics. Work in this area has… provided mutants, produced practical procedures to simplify the manipulation of viral genes, and generated information about the molecular architecture and organization of genes characteristic of pox viruses. One of the most intensively studied regions of the viral genome is the HindIII D region of vaccinia, in which a combination of classical and molecular genetic analysis of the region has been particularly revealing. Within this region are open reading frames, some of which are expressed early and others late, organized in a fashion which is now known to be typical of these viruses. Other studies, related to temperature sensitive, drug resistant, and drug dependent mutants, are also discussed. Each of the other reviews included in this volume summarizes areas of research which have depended heavily on the genetics of the system. The intracellular site of a poxvirus infection is mostly, if not exclusively, limited to the cytoplasm which dictates several interesting biological ramifications. For example, poxvirus transcription must occur in the cytoplasm, rather than in the nucleus. The virus copes with this situation by incorporating into the virion the enzymatic machinery necessary to initiate transcription from input virus.

In the more than 200 years since that time we have seen the remarkable eradication of smallpox. It is difficult to overstate the signi…ficance of that achievement. It not only removed a disease that must rate as one of humankind's greatest scourges, but also demonstrated the effectiveness of the general principle of vacci- tion in our battles against disease. This book begins with a review of smallpox and its causative agent, Variola virus. The vaccine used in the successful smallpox eradication c- paign, vaccinia virus, is reviewed in the following chapter that describes its origin and its use as a vaccine, as well as the current understanding of the molecular biology and pathogenesis of this virus. Vaccinia virus is the most intensively studied poxvirus and the descriptions of the biology of this virus are relevant to all vertebrate poxviruses. The eradication ofsmallpox has drawn attention to the potential threat posed by other orthopoxviruses that infect humans, particularly Monkeypox virus. A description of this virus is given in the third chapter. Jenner's ori- nal vaccine is believed to have been Cowpox virus and this virus is reviewed in the chapter by Essbauer and Meyer.
