Taschenbuch. Zustand: Neu. Druck auf Anfrage Neuware - Printed after ordering - Genomic instability is a major threat to living organisms. To counteract the damaging effects posed by endogenous and environmental agents, such as chemicals or radiation, micro-organisms devote several percent of their genome to encode proteins that function in the repair and recombination of DNA. For many years, a relatively small group of scientists have carefully delineated the molecular mechanisms of these repair processes, using the simplest model systems available, namely Escherichia coli and Saccharomyces cerevisiae. These studies, which until recently had only moderate impact outside of the field, now provide the cornerstone for exciting new research into analogous processes in human cells. The reason for this is the revelation that the biochemical pathways for the accurate replication, repair and recombination of DNA have been conserved through evolution. New research shows that human cells use DNA repair mechanisms that are analogous to those found in bacteria to overcome the damaging effects of environmental mutagens and carcinogens. Of particular significance is the observation that certain cancers are caused by defects in DNA repair enzymes. For example, the human inherited non-polyposis colon cancer is now known to be caused by defects in the enzymes that repair DNA mismatches. Because much detailed information has been accumulated in studies of analogous mismatch repair proteins from bacteria, rapid progress can now be made.

Zustand: Sehr gut. Gebraucht - Sehr gut -Recombination mechanisms allow cells to break and reattach DNA sequences. They are important for repair of damaged DNA. They also help generate additional genetic diversity in sperm and eggs cells by mixing DNA sequences from maternal and paternal chromosomes. This volume explores the molecular details of recombination and their roles in genome maintenance and meiosis. Gebundene Ausgabe, Größe: 23.2 x 16.7 x 2.8 cm.

Zustand: Sehr gut. Zustand: Sehr gut | Seiten: 580 | Sprache: Englisch | Produktart: Bücher | Homologous recombination is important in various aspects of DNA metabolism, including damage repair, replication, telomere maintenance, and meiosis, and yeast genetics has successfully provided a framework for the mechanism of homologous recombination. Divided into four convenient sections, DNA Recombination: Methods and Protocols covers recent techniques that best utilize the advantages of the yeast system, prescribing to the belief that yeast will keep serving as a great model organism to study homologous recombination. Chapters have also been included for such exceptions as the group of genes involved in recombination that are found solely in higher eukaryotes, such as BRCA2. And looking forward, a necessary step in the direction of understanding the homologous recombination process is to isolate the machine and let it work in a test tube. Understanding the design by studying the appearance and behavior of the machinery as a single molecule will be an important milestone toward understanding the mechanism of action of the machinery. Techniques covering these topics have also been included. Written in the successful Methods in Molecular Biology¿ series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible protocols, and notes on troubleshooting and avoiding known pitfalls. Authoritative and easily accessible, DNA Recombination: Methods and Protocols serves as an ideal guide to scientists of all backgrounds with its well-honed methodologies and strives to bring the reader to the next level of understanding regarding this vital subject.
