Neuware -Die protektive Wirkung von Immunsuppressiva auf den hepatischen Ischämie-Reperfusionsschaden (I/R-Schaden) deutet darauf hin, dass T-Zellen bei diesem alloantigen-unabhängigen Ereignis eine Rolle spielen. Die Mechanismen der Aktivierung von CD4+ T-Zellen bei alloantigen-unabhängiger I/R der Leber sind jedoch weitgehend ungeklärt. In der aktuellen Arbeit wurde daher die Rolle von CD4+ T-Zellen während hepatischer I/R in vivo analysiert. Erstmals konnte über die Art, die Kinetik und die Lokalisation von CD4+ T-Zell-Endothelzell-Interaktionen Auskunft gegeben werden. CD4+ T-Zellen interagieren mit Thrombozyten während der hepatischen I/R und die Ausbildung des hepatischen I/R-Schadens wird über kostimulatorische Signalwege, wie CD40-CD40L und CD28-B7, moduliert. Dieses findet MHC-Klasse II-unabhängig statt. Des Weiteren konnte in diesen Untersuchungen erstmals gezeigt werden, dass eine reziproke Aktivierung von CD4+ T-Zellen und Kupffer-Zellen während hepatischer I/R stattfindet. Die aktuell gewonnenen Daten unterstreichen die Notwendigkeit weiterer Untersuchungen zur Optimierung immunsuppressiver Therapien bzw. zur Etablierung Toleranz-induzierender Pharmazeutika.Books on Demand GmbH, Überseering 33, 22297 Hamburg 104 pp. Deutsch.
