Oxidative stress (OS) is associated with many chronic states including cancer. Curcumin, a bioactive chemical of turmeric has potent antioxidant effects. We utilized metabolomics to evaluate signatures of OS in BOP (N-nitrosobis-2-(oxopropyl) amine) induced hamsters and their effect by curcumin. 32 hamsters were randomized into treatment (n=20; OS induced by BOP) and control (n=12) groups. After 12 weeks each group was subdivided by no (T0, C0) or 2% curcumin (T2, C2) diet. Proton nuclear magnetic resonance spectroscopy was conducted on urine and analyzed using Principal Component Analysis PCA;SIMCA P+, Umetrics. The PCA score plots showed clear separation between spectra from treatment (T0) and control (C0) groups, and curcumin fed (T2) groups (p<0.05). Partial Least Square (PLS) analysis correlated the metabolomic profiles with Total Antioxidant, cyclooxygenase-2 and NF-kB activity well. Metabolites responsible for the inter-group differences were linked to the pyrimidine, purine and glycine, serine and threonine pathways. With further investigation these metabolites, in addition to providing a mechanistic insight, may serve as potential signatures of OS.
