This book presents protocols for 3D tissue engineering, encompassing biochemical, immunostaining, and gene expression methods for the fabrication of scaffolds from synthetic polymers, peptides that self-assemble, and quantitative methods for characterizing the mechanical and chemical properties of these scaffolds. Also covered is the combination of scaffolds, hydrogels, and cells as building blocks to create even larger structures using layer-by-layer deposition or macromolding approaches. Several chapters are focused on the challenges of specific tissues and organs, such as cartilage and the nervous system. Each protocol includes an abstract, a list of key terms, a detailed introduction on scientific background and practical issues, and a list of materials, then details step-by-step methods, with notes on data analysis, interpretation, and application. Francois Berthiaume is an associate professor in the Department of Biomedical Engineering at Rutgers University. He holds a B.S. and a Ph.D. in chemical engineering from Laval University and The Pennsylvania State University, respectively.
Jeffrey Morgan is an associate professor in the Department of Molecular Pharmacology, Physiology and Biotechnology at Brown University. He holds a B.S. from Syracuse University and a Ph.D. from Harvard University.
